The Wax Secretion Characters and the Content Changes of FAS and FAE in the Different Stages of Matsucoccus matsumurae (Kuwana)

Abstract:To study the wax secretion stages of Matsucoccus matsumurae (Kuwana) and the roles of fatty acid synthase (FAS) and fatty acid elongase (FAE) during these stages, microscopic observation techniques and enzyme content determination method were used. The result showed that the secretion of wax in male scale insects mainly happened in the 2nd instar nymphs and 3rd instar nymphs. The “pearl-shaped” 2nd instar nymphs would secret less amount of wet wax to form a thin layer on the body surface, and the wax filaments was secreted by the spiracles located on the lateral body. The 3rd instar nymphs secreted a large amout of wax filaments to form cocoons. The females would secret wax in the 2nd instar nymphs and adult stages. The variation of FAS contents was closely related to the wax-secretion activity of scale insect. In the developmental stage of males, the highest content of FAS was in the 3rd instar nymph stage, followed by the 2nd instar nymph stage, and the lowest in the pupal stage. The highest contents of FAS were observed in the early and medium wax secretion stages of adult females. The content of FAS would decrease to the lowest level when the egg deposition was finished. The middle value of FAS was observed in the 2nd instar nymph stage. The variation of FAE contents was similar to that of FAS except the 2nd instar nymph stage. These results indicated that FAS and FAE may play an important role in the synthesis and secretion processes of wax.
