Random Amplified Polymorphic DNA Analysis (RAPD) of Misgurnus anguillicaudatus and Paramisgurnus dabryanus from Nansi Lake

Abstract:The genetic differentiation of Misgurnus anguillicaudatus and Paramisgurnus dabryanus from Nansi Lake, Shandong Province was studied by RAPD. Totally 243 amplified DNA fragments (2003000bp) were obtained by using 5 random oligonucleotide primers from 7 samples of the two kinds of fish. Of them 4 fragments were shared by the 7 samples, 18 fragments by 3 samples of Paramisgurnus dabryanus and 12 fragments by 4 samples of Misgurnus anguillicaudatus. RAPD phenogeam was constructed by UPGMA using genetic distance of samples calculated by PoP-Gen32 software. It has been suggested that the 7 samples were divided into 2 clusters, which is consistent with the results based on morphological study in which they belong to Misgurnus anguillicaudatus and Paramisgurnus dabryanus.
