Analysis of DAPI Karyotype of Bighead Carp (Aristichthys nobilis)

Abstract:The Aristichthys nobilis chromosomes were prepared by kidney cells and stained using fluorescence dye DAPI (4’, 6’-diamidino-2-phenylindole). Then the daryotype and the distribution of chromatin of Aristichthys nobilis were studied. The results showed that the chromosome number of diploid was 48 and the karyotype formula is 30M+14SM+2ST+2T. The well-proportioned fluorescence brightness was detected in interphase nucleus, which indicated that the heterochromatin distribution in interphase nucleus was relatively symmetrical. But the bright regions lied on the short arm of chromosome 1 and 4, and they distributed at the centromere of the other chromosomes. These revealed that heterochromatin was mainly concentrated on the short arm of chromosome 1 and 4, while mainly distributed at the centromere of the other chromosomes.
