Expression of PI3K and Akt Protein in Rat Cardiac Hypertrophy

Abstract:Objective To provide morphological data for the investigation of the cardiac hypertrophy signal transduction mechanism and reversal of cardiac hypertrophy by researching the expression of PI3K and Akt in cardiac tissue in isoproterenol-induced myocardial hypertrophy. Methods Twenty healthy adult SD rats were randomly allocated into 2 groups: experimental group (n=20) and control group (n=10). The heartswere taken from rats after a week, paraffin sections of rat myocardial tissue were made in order to stain by HE for pathological analysis and measure the contents of index of myocardial hypertrophy; The expression and distribution of p-PI3K, p-Akt protein in the experimental group and control group were detected by immunohistochemistry and immunofluorescence. Results Index of myocardial hypertrophy in the experimental group were significantly higher than those of the control group; the results of immunohistochemical detection showed that in protein expressions area, average optical density of p-PI3K, p-Akt in the experimental group were significantly higher than those in the control group. Compared with thecontrol group, the positive expression of p-PI3K and p-Akt in the experimental group was significantly increased. Conclusion A model of cardiac hypertrophy can constructed by the method of continually injecting low-dose ISO to rats. p-PI3K and p-Akt protein expression are related to the occurrence of cardiac hypertrophy and relevant development; the PI3K/Akt signaling pathway activation could be one of the mechanisms of cardiac hypertrophy.
