Characteristics of Pathological Changes in Chronic Obstructive Pulmonary Disease (COPD) Rat Model

Objective To study the expression of pulmonary surface-active substance related protein A (Surfactant protein A, SP-A) of chronicobstructive pulmonary disease (COPD) rat model and ultrastructural changes in the lung tissue of COPD. Methods The rat model of COPD was established by both intratracheal instillation of lipopolysaccharide (LPS) and passive smoking for 28 days. Observe the general condition of rats, test the expression level of Surfactant protein A (SP-A) by immunohistochemistry, and observe the ultrastructural changes of alveolar type epithelial cells through transmission electron microscope. Results Compared with the blank group, the average numbers of SP-A-positive cells per alveolus in other groups showed significant differences (P<0.01). Conclusion It is reliable that the rat model of COPD was established by the combination of intratracheal instillation of lipopolysaccharide (LPS) and passive smoking for 28 days, and the model was still stable after ending the combination two weeks later.
