Spatiotemporal Expression of Tumor Metastasis Inhibitory Factor CD63/ME491 Gene in Embryo and Delayed Implantation Uteri in Mice

Object To identify mRNA and protein expression of tumor metastasis suppressor gene CD63/ME491 in embryo and delayed implantation uteriin mice. Methods mRNA and protein expression of CD63/ME491 were detected by reverse transcription polymerase chain reaction, immunohistochemistry and immunofluorescence. Results 1.mRNA and protein expressions of ME491/CD63 were detected in all stages of embryo development. ME491/CD63 mRNA was dramatically abundant in morula and blastocyst. Its protein was detected in the cellular membrane and cytoplasm of embryos. 2. In uteri of delayed implantation mice, expression of CD63/ME491 mRNA was detected from D5 to D8, and its quantity gradually decreased, but it remarkably increased after estradiol activation (P<0.05). Expression of CD63/ME491 protein was weakly positive in stromal cells underneath epithelial cells on D5, no expression was detected on D6~D8, however, there was strong positive expression in stromal cells underneath epithelial cells after estradiol activation. Conclusions 1. Dynamic expression of CD63/ ME491 in mouse embryo showed it might play roles in embryonic development. 2. Expression of CD63/ME491 in mouse uterus might be regulated by estrogen.
