Primary Culture and Identification of the Olfactory Ensheathing Cell and Olfactory Nerve Fibroblast from Adult Rats

Abstract: objective To establish a method or procedure for obtaining primary cultures of olfactory ensheathing cells and olfactory nerve fibroblasts from the adult rat olfactory bulb. Methods Olfactory ensheathing cells and olfactory nerve fibroblasts were isolated and cultured from the outer olfactory nerve fiber and glomerular layers of 2.5 month adult SD rat olfactory bulbs, but without purification. On the 7th, 10th, and 14th days, the cultured cells were identified by immunocytochemistry and the purity of the olfactory ensheathing cell was calculated. Results The cultured olfactory ensheathing cell presented bipolar or multipolar morphology and was p75 NGFR immunopositive; the olfactory nerve fibroblast has a flattened fibroblast-like morphology and was fibronection immunopositive; both of two cells were found to be vimentin immunopotive. The purity of the olfactory ensheathing cell was 34.1%, 25.6%, 8.6% on days 7, 10, and 14 respectively. Conclusion The cultures from the outer two lays of adult rat olfactory bulb mainly contain olfactory ensheathing cell and olfactory nerve fibroblast. The percentages of olfactory ensheathing cell in culture gradually decrease with extension of culture times.
