Species Authentication of Bama Miniature Pig through Inter-simple Sequence Repeat

Abstract:In this study, species identification of Bama miniature pig was carried out by DNA fingerprinting of meats from Tibetan pig, Huanjiang miniature pig, Bama native pig, and Chenghua pig through inter-simple sequence repeat (ISSR) and mitochondrial DNA (mtDNA) analysis. One clear and repeatable primer pair named No. 53 was selected from 55 primers, which successfully separated Bama miniature pig from the other four pigs. To further differentiate Bama miniature pig and Tibetan pig, specific tandem repeat sequence of Tibetan pig ‘TGTGTACGCA’ was found out in the mtDNA. According to the results of ISSR and UPGMA cluster analysis, primer No. 53 was able to attribute to the identification of the five different pigs and one unknown pig. Therefore, ISSR coupled with UPGMA and mtDNA analysis represented an effective tool in species identification.
