Influence of the Ovary Size to Oocyte in vitro Mature in Goat

Abstract:Objective The influence of ovary size on the in vitro maturity of oocyte was investigated. Method The ovaries of goat from slaughter house were divided into three groups according to the weight (groupⅠ<0.95 g; group Ⅱ, 0.95~1.7 g; group Ⅲ>1.7 g). Some statistic indices including maturation rate, parthenogenetic cleavage rate, parthenogenetic blastocyst rate, cloned embryo cleavage rate and cloned embryo blastocyst rate of the three groups were determined in vitro. Result The in vitro maturation rates of oocytes from different groups were 62.25%, 43.58% and 40.7%, respectively. Parthenogenetic cleavage rates of oocytes from different groups were 83.77%, 82.92% and 79.73%, respectively. Parthenogenetic blastocyst rates of oocytes from different groups were 19.51%, 18.9% and 18.78%, respectively. Cleavage rates of cloned embryo reconstructed using oocytes from the three groups by handmade cloning method were 67.36%, 65.97% and 66.33%, respectively, and the blastocyst rates of the cloned embryos were 11.63%, 13.41% and 12.29%, respectively. Oocyte maturation rate of groupⅠwas significantly higher than that of others. Moreover, no significant difference of parthenogenetic cleavage rate, parthenogenetic blastocyst rate, cleavage rate of cloned embryos and blastocyst rate of cloned embryos was observed among the three. Conclusion The results showed that the in vitro maturation rate of oocytes of small size ovaries was higher than that of the big ones. The size of ovary only affects oocyte maturation rate rather than developmental capacity.
