Effect of Arecoline on the ERα and PR of Pregnant Mice Uterus

Abstract:ICR pregnant mice were used to explore the antifertility effects of arecoline and its influence on the ERα and PR. The pregnant mice were intraperitoneally injected with 0 mg/kg·day, 5 mg/kg·day, 10 mg/kg·day and 20 mg/kg·day arecoline. The statistical results showed that arecoline decreased the number of embryo implantation and uterine weight in a dose-dependent manner. When the dose was 20 mg/kg·day, the number of embryo implantation was decreased to 59% and the uterine weight was decreased to 77% in arecoline-treated group, in compared to the control. The result of HE staining indicated that the 20 mg/kg·day arecoline treated mice showed an opened endometrial lumen on D5 and failed in decidualization on D6. Additionally, the results of real-time PCR and immunohistochemistry demonstrated that the both mRNA and protein of ERα and PR in uterus were significantly up-regulated during the implanting period (P<0.05).
