Isolation and Culture of Hepatocytes from Regenerating Liver of Rat

Abstract:Hepatocytes of rat treated by partial hepatectomy after 0 h, 12 h, 12 h, 24 h, 48 h, 72 h, 120 h, and 168 h were isolated and purified by collagenase digestion and density gradient centrifugation. The viability of regenerated hepatocytes was over 95%, and the purity was more than 96%. The regenerated hepatocytes grow well in DMEM containing 10% fetal calf serum, and were capable of secreting albumin and synthesizing urea. DNA synthesis of hepatocytes was initiated after 24 h and achieved the peak at 72 h.
