Characterization of GPR15 in Chickens: Cloning, Tissue distribution and Functional analysis

Abstract:In this study, the cDNA sequence of chicken GPR15 was cloned using the cDNA derived from the chicken's colon. The sequence length of cDNA coding region of chicken GPR15 is 1107bp, which consists of 1 exon and encodes a protein containing 368 amino acids. Amino acid sequence analysis showed that chicken GPR15 has a classical seven-transmembrane structure of GPCR. Synteny analysis showed that GPR15 of chicken is the orthologous gene of GPR15 of zebra finch, Chinese turtle, lizard, and human GPR15; Monitored by the pGL3-CRE-luciferase reporter system and pGL4-SRE- luciferase reporter system, GPR15 can’t be activated by the endogenic ligand of chicken APJ (Apelin and Apela) in CHO cells. The tissue expression pattern showed that chicken GPR15 is highly expressed in the spleen, and there is a certain amount of expression of GPR15 in caecum and lung. These results laid a foundation for exploring the functional properties of GPR15 and the physiological functions of GPR15 in chickens.
