The investigation of cannabinoid receptor 1 on lipid metabolism in diet induced obese mice

Abstract:Abstract: To study the regulation mechanism of CB1 on lipid metabolism in diet induced obese mice. The C57BL/6J male mice were induced with high-fat diet, and then the effect of rimonabant on the body weight, liver weight and serum biochemical index of mice were detected. In particular, the CB1 and CPT1 mRNA levels, and CB1 protein levels in subcutaneous fat, visceral fat, skeletal muscle, liver and heart were also tested. The results showed the body weight, liver weight and the mRNA and protein levels of CB1 were decreased by rimonabant significantly (P<0.05). Simultaneously, the levels of TC, HDL, adiponectin and leptin were improved (P<0.05). Furthermore, the mRNA levels of CPT1A and CPT1B in each tissue were increased, while CPT1A and CPT1B were not detected obviously in the heart. In conclusion, we confirmed that CB1 can play a role in regulating lipid metabolism through controlling of CPT1A, CPT1B genes. Our paper provides a reliable basis for clinical treatment of lipid metabolism disorders.
