Gene cloning, molecular evolution and tissue expression of G protein-coupled receptor 161 in chicken

Abstract:G protein-coupled receptor 161 (GPR161), as a member of GPCR superfamily, is an orphan receptor playing important role in lens development and neurulation. Owing to its shared structure with scaffolding proteins, GPR161 draws wide attention for its potential in signal transduction. Using chicken as experimental model, the cDNA sequence, molecular evolution and tissue expression of chicken GPR161 gene were reported in the present study. The full-length coding sequence of chicken GPR161 gene is 1566 bp in length encoding 521 amino acids. Sequence analysis revealed that GPR161 of chicken shares high sequence similarity with that of human (83.0%), mouse (82.6%) or zebrafish (65.8%). Phylogenetic analysis of GPR161 shows that relations between chicken and human or mouse are closer than that between chicken and zebrafish. By real-time quantitative PCR, the mRNA of GPR161 is expressed with high abundance in testis or ovary, brain, heart and muscle. Our data provides basis to elucidate the physiological roles of GPR161 in birds.
