Influence of Americana Periplaneta Extract on Antioxidant Index in Blood and Ovary of Rats

Abstract：To study the effect of Americana periplaneta extract on antioxidant index of blood and ovary in rats through this experiment. Methods: The 17 adult female SD rats were used, and were randomly assigned to three groups after a week of adaption to the environment. The control group of 5 rats were only filled with physiological saline (9 ml/kg) through stomach perfusion. The 6 rats were used respectively in Low dose group (9 ml/kg，0.03g/ml) and High dose (9 ml/kg，0.09 g/ml) which were filled with periplaneta Americana extract through stomach perfusion. Rats were treated every other day for 30 days. After the experiment, the blood and ovaries of rats were collected, and the content of malondialdehyde (MDA), superoxide dismutase (SOD), catalase (CAT), nitric oxide(NO), glutathione (GSH-PX) in the serum, the gene expression of SOD, GSH-PX and CAT in the ovary were detected. Results: Compared with control group, high dose group (p = 0.085) and low dose group (p = 0.091) have a significant trend to reduce NO level in serum; High dose and low dose of periplaneta Americana extract increased significantly the content of SOD in serum of rats (p &amp;amp;lt; 0.05). However, high dose group decreased significantly the content of MDA in serum (p &amp;amp;lt; 0.05). In addition, the high dose and low dose group had higher ovarian expression of SOD gene (p &amp;amp;lt; 0.05). Conclusion: Americana Periplaneta extract increased significantly the content of SOD as the antioxidant index in the serum of these rats and reduced significantly the oxidative stress indicators of NO and MDA content. It also increased significantly the expression of SOD gene in ovary. It indicated that Americana Periplaneta extract has a certain antioxidant stress function to the ovary of rats.
