基于RNA-seq分析美洲大蠊提取物对结直肠癌细胞信号通路的影响

中文摘要:美洲大蠊Periplaneta americana提取物能够抑制多种肿瘤细胞生长,甚至能使肿瘤细胞发生凋亡,但是其对结直肠癌细胞信号通路的影响尚不清楚。本文基于RNA-seq技术初步分析了美洲大蠊提取物联合顺铂处理结直肠癌细胞后,细胞内信号通路的变化。结果表明:30 μM顺铂和0.6%康复新液联合处理组与对照组相比,共有1 901个差异基因,其中1 555个基因表达上调,346个基因表达下调;30 μM顺铂和0.8%精粉酵解液联合处理组与对照组比,共有2 587个差异基因,其中2 183个基因表达上调,404个基因表达下调;30 μM顺铂和100 μg·mL-1精粉联合处理组与对照组相比,共有1 488个差异基因,其中1 164个基因表达上调,324个基因表达下调。用WEGO和DAVID进行差异基因的GO和KEGG富集分析发现,美洲大蠊提取物联合顺铂处理组与对照组相比,表达上调的差异基因主要富集在p53、细胞粘附分子、MAPK、细胞凋亡等信号通路;表达下调的差异基因主要富集在氨基酰-tRNA生物合成、氨基酸生物合成、抗生素生物合成、一碳代谢等信号通路。
英文摘要:Periplaneta americana extracts can inhibit the growth of several tumors, and even induce tumor cell apoptosis. However, the influence of P. americana extracts on the signaling pathways in cancer cells is still unknown. In this study, RNA-seq method was used to investigate the changes of signaling pathways in colon cancer cells that treated with P. americana extracts and cisplatin. The results showed that, comparing to the untreated group, colon cancer cells that treated with 30 μM cisplatin and 0.6% KFXY had 1 901 differentially expressed genes (DEGs), including 1 555 up-regulated genes and 346 down-regulated; 30 μM cisplatin and 0.8% JFJJ treated group had 2 587 DEGs, including 2 183 up-regulated genes and 404 down-regulated; 30 μM cisplatin and 100 μg·mL-1 JF treated group had 1 488 DEGs, including 1 164 up-regulated genes and 324 down-regulated. GO analysis and KEGG pathway analysis showed that the up-regulated DEGs were enriched in p53 signaling pathway, cell adhesion and apoptosis, while the down-regulated DEGs were enriched in aminoacyl-tRNA biosynthesis, one carbon pool and biosynthesis of amino acids.
