中文摘要:利用阿利新蓝和过碘酸雪夫氏(AB-PAS)染色方法,研究了邻苯二甲酸二乙基己酯(DEHP)对鲤鱼鳃及消化道粘液细胞密度的影响,实验分为空白对照组、吐温80组及5 mg/L20 mg/L80 mg/L160 mg/L DEHP组,染毒20 d结果表明,吐温80组与空白对照组相比差异不明显。5 mg/L组口腔上皮处粘液细胞密度显著低于吐温80组外,在实验剂量范围内,鳃丝、口腔上皮、前肠、后肠粘液细胞密度均极显著低于吐温80组。中肠处粘液细胞密度5 mg/L20 mg/L组与吐温80组相比差异不显著,80 mg/L组显著低于吐温80组,160 mg/L组极显著低于吐温80组。鳃丝、口腔上皮、中肠处粘液细胞密度均随DEHP浓度的增大而减小,而前肠和后肠在80 mg/L组粘液细胞密度最小,降幅最大。实验结果还表明:在实验剂量范围内,与吐温80组相比,DEHP能降低鳃丝、口腔上皮处型,前肠型,中肠型,后肠型粘液细胞密度。DEHP能提高鳃丝型,口腔上皮和前肠型,中肠型和后肠型粘液细胞密度。DEHP能减小鳃丝及消化道粘液细胞的密度,对各类型粘液细胞密度的影响因部位不同、DEHP浓度不同而异。
英文摘要:The mucous cell types of digestive tract and gill of Cyprinus carpio Linnaeus were classed by alcian blue and periodic acid schiff staining (AB-PAS), and the effect of di-(2-ethylhexyl) phthalate (DEHP) on mucous cell density of these tissues were studied. Six groups including blank control, tween-80 control, and 5 mg/L, 20 mg/L, 80 mg/L, 160 mg/L DEHP groups treaded for 20 days were used. The result showed that there was no significant difference between the tween-80 control and the blank control. In the range of tested experimental doses, the mucous cell densities of gill filament, oral epithelium, foregut and hindgut were decreased significantly compared with the tween-80 control group (P<0.05). The mucous cell densities of gill filament, oral epithelium and middle intestines were decreased with the increasing of the concentration of DEHP, and the lowest densities were observed in foregut and hindgut of 80 mg/L DEHP treated group. It also showed that after immersing stimulated with DEHP, the densities of type Ⅱand mucous cell in oral epithelium, gill filament, type Ⅰ, Ⅱ, Ⅳ in former intestines, type Ⅰ, Ⅲ, Ⅳ in middle intestines and type Ⅱ, Ⅲ in retrial intestines were all decreased. However, the densities of type Ⅰ, Ⅲ mucous cell in gill filament, type in oral epithelium and former intestines, type Ⅱ in middle intestines and type in retrial intestines were increased. In conclusion, DEHP can reduce the density of mucous cells in digestive tract and gill filament. The effect of DEHP on the density of mucous cells is correlated with the part of digestive tract and concentration of DEHP.
