成年乌金猪、青峪猪和成华猪免疫器官组织TLRs和抗菌肽基因表达比较研究

中文摘要:TLRs和抗菌肽是机体天然免疫系统的重要组成部分,是抵御外界有害微生物或病毒入侵的重要屏障。成华猪、乌金猪、青峪猪是中国优良地方猪种,具有繁殖力高、抗病耐逆性强等优点。本实验比较了8月龄成年乌金猪、青峪猪、成华猪与约克夏猪的消化道、呼吸道局部免疫组织和全身免疫器官的先天免疫特点,采用实时荧光定量PCR技术检测TLRs(TLR1、TLR2、TLR4、TLR7、TLR9)和抗菌肽(pBD-1、PR-39)基因在4种猪的胸腺、脾脏、扁桃体、肠系膜淋巴结和肺门淋巴结中的表达差异。结果显示,与其他猪种相比,成华猪胸腺中TLR-7、PR-39,脾脏中TLR4、TLR7,肠系膜淋巴结中TLR4、TLR9,肺门淋巴结中TLR1、TLR4、TLR7、TLR9 mRNA表达量高(P <0.05);乌金猪胸腺中TLR1、TLR9,扁桃体中TLR1、TLR2、TLR4、TLR7、TLR9、pBD-1,肠系膜淋巴结中TLR7mRNA表达量高(P <0.05);青峪猪扁桃体和肠系膜淋巴结中PR-39 mRNA表达水平高于其他猪种(P <0.05)。这些结果为进一步利用优良地方猪杂交育种和筛选抗病分子标记提供了新的线索。
英文摘要:Toll-like receptors (TLRs) and antimicrobial peptides are important components of innate immunity of the natural immune system and an important barrier to protect against harmful microorganisms or viruses. Chenghua pig, Wujin pig and Qingyu pig are well known Chinese unique local subspecies of pig breed, and they own the merits of high fertility and strong disease resistance. In this study, we compared the innate immunity characteristics of 8-month-old Wujin pigs, Qingyu pigs, Chenghua pigs and Yorkshire pigs with local immune tissues and systemic immune organs. Real-time fluorescent quantitative PCR was used to detect the expressions of TLRs (TLR2, TLR4, TLR7 and TLR9) and antimicrobial peptides (pBD-1 and PR-39) genes in thymus, spleen, amygdala, mesenteric lymph node and hilar lymph nodes. The results showed that compared to the control pigs,the expression levels of TLR7 and PR-39 in thymus, TLR4 and TLR7 in spleen, TLR4 and TLR9 in mesenteric lymph nodes, and TLR1, TLR4, TLR7 and TLR9 mRNA in hilar lymph nodes of Chenghua pigs were significantly higher (P<0.05); The expression level of TLR1, TLR9 in the thymus, TLR2, TLR4, TLR7, TLR9 and pBD-1 in the tonsils, TLR7 in the mesenteric lymph nodes of Wujin pig were significantly higher (P<0.05); The expression level of PR-39 mRNA in the tonsils and mesenteric lymph nodes of Qingyu pigs was higher than other pig breeds (P<0.05). These results provide useful clues for screening of disease-resistant molecular markers of local pig hybrid breeding in the future.
